Subculture: | seed out at ca. 0.3-0.5 x 106 cells/ml; maintain at ca. 1.0 x 106 cells/ml; split saturated culture 1:3 to 1:8 every 2-5 days. Upon cytokine withdrawal cells do not die but undergo a stationary survival status. CAVEAT: by favoring selective growth, suboptimal culture of cytokine-dependent cell lines may promote outgrowth of factor-independent subclones. Selective conditions include cytokine insufficiency and inadequate cell density. Thus the DSMZ cannot guarantee indefinite stability of factor-dependence of cell lines. |
