At the DSMZ the cyanobacterial cultures are usually cultivated as a standing culture in 25 cm2 plastic tissue culture flasks with filter screw caps, which are routinely filled with 20 ml of fresh medium each. For subcultivation, scrape off the attached material with a disposable pipette and crush it by pumping it up and down several times or crushing it with the pipette tip. Then soak up 2-3 ml of biomass and transfer to the fresh bottle. Light cycle: 16 hours of illumination at 300-350 lux and 8 hours dark period. Transfer interval of 12 weeks.
