Subculture: | split saturated culture 1:2 to 1:4 every 2 days; seed out at 0.3 x 106 cells/ml; maintain at 0.2-0.4 x 106 cells/ml; best to culture cells for the first 5-7 days in a 12-well-plate with HFCS, then cells may be transfered to flasks; after 14 days, cells become slightly adherent (about two-thirds of them), but can be detached by slapping the flask; the HFCS concentration can be decreased after the initial 2 weeks to about 50% (1:100) or 25% (1:200) |
