Origin: | derivative of 293T (HEK293T), established in 2021 by CRISPR/Cas9-mediated modification of the CMP-sialic acid transporter-encoding gene SLC35A1 resulting in decreased/absent levels of the protein. The cells are described to harbour a functional knockout of the SLC35A1 gene leading to increased susceptibility to transduction with adeno-associated virus 9. The cells are therefore suited for AAV9 transduction potency assays |

Risk assessment: | The parental cell line 293T was established by transfection of the cells with sheared DNA of adenovirus type 5. The cells contain 4 to 5 copies of the left end of the virus (12% of the viral genome) including E1a and E1b genes and one copy of the right end of the virus (10% of the genome) including the E4 gene. No active viruses are produced. The cells are classified biosafety level 1 and the genetic modification renders the cell line level S1 according to German genetic engineering act. |
