Subculture: | seed out at ca. 0.8 x 106 cells/ml in 24-well plates; maintain at ca. 0.5-1.0 x 106 cells/ml; split culture 1:2 once or twice a week; after thawing cells may need some days to recover and a certain amount of cell debris may be allways visible; CAVEAT: by favoring selective growth, suboptimal culture of cytokine-dependent cell lines may promote outgrowth of factor-independent subclones. Selective conditions include cytokine insufficiency and inadequate cell density. Thus, the DSMZ cannot guarantee indefinite stability of factor-dependence of cell lines. |
