Remarks: | Genomic DNA (a 2.4 kb and a 0.9 kb PstI fragment) of Ralstonia eutropha HF39 was cloned into the 6.295 kb vector pSK(BEC/pp:3,3) which is a derivative of pBluescript SK- and transformed into the host E. coli XL1-Blue. On complex medium the cells produce a blue pigment characterised as indigo by several methods. The 1.251 kb ORF (bec) was cloned and sequenced. The gene product catalyzed formation of indoxyl, a reactive preliminary stage for indigo production. Strain forms long cells and colonies get an indigo blue colour after prolonged incubation. |
