Remarks: | Bioluminescent reporter plasmid for naphthalene catabolism, developed by insertion of Tn4431 of the lux gene cassette from Vibrio fischeri into a naphthalene catabolic plasmid in P. fluorescens. Insertion site of the lux transposon was the nahG gene encoding for salicylate hydroxylase. Luciferase-mediated light production from P. fluorescens strains harbouring this plasmid is induced on exposure to naphthalene or the regulatory inducer metabolite salicylate. In continuous culture, rapid light production occurs after 15 minutes. This strain does not mineralize naphthalene completely and accumulates salicylate, due to insertion in the lower pathway operon. The lux transposon is located on a 1.6 kb PstI fragment adjacent to the nahH gene. |
